- Ever wondered what happens if you remove Tfh cells from established germinal centres?
- We did. We wanted to disentangle the role of Tfh cells in GC formation from their ongoing maintenance, so we used a Il21cre iDTR approach to delete Tfh cells from established GCs. Our hypothesis was that the GC would collapse, but it was surprisingly resilient.
- The GCs contracted, because of the lack of Tfh cells to support ongoing B cell proliferation, but then recovered when Tfh cells repopulated the GC. Resulting normal output and affinity maturation.

- Lung germinal centres are smaller and less densely packed with cells that those in lymph nodes, but despite this @sanmarti92.bsky.social @smguillaume.bsky.social and @wsfoster.bsky.social were able to show they still showed light and dark zone polarisation.
- @smguillaume.bsky.social used single cell sequencing to show that after NP-KLH/house dust mite treatment, lung germinal centre B cells were able to acquire high affinity B cells, although this was not as fast as in the lymph node germinal centres.

View full threadFrom analysis of clones from the lung and the lymph node, it was clear that lung B cells may directly seed local lung memory B cell responses, which @smguillaume.bsky.social was able to show in vivo by blocking lymphocyte egress from lymph nodes with FTY720.
