- Reposted by Luca Giorgetti lab @FMI(23) If you move SRR2 further than ~20kb from the Sox2 promoter, it cannot support cohesin-independent action of the distal enhancer. ~20kb was the cuttof we say at Car2 for cohesin-independent action of enhancers. Makes sense!

We show how loop-extruding SMC complexes can maintain DNA entrapment while bypassing obstacles on DNA — including transcription machinery & potentially other SMCs. - 2/ We started by asking: How often, and for how long, do two genomic sequences meet in the cell nucleus within an arbitrary encounter radius? And how does this depend on the loop extrusion activity of cohesin?
- 3/ Previously, @andersshansen.bsky.social's lab and us (@piamach.bsky.social @pavelkos.bsky.social ) showed that CTCF loops last ~10-30 minutes. But how about any other encounters, i.e. those that are not stabilized by CTCF?
