- Question for fellow #mitochondriacs on #MitoBsky - what are your preferred/favorite/best methods to isolate mitochondria from mouse tissues? shearing + UC? nitrogen cavitation + UC? antibodies (anti-Tom22?)? Miltenyi + antibody? other combinations? bonus if the tissue is brain 🙃 #mitochondria

- I tried that before with mixed success… problems were: 1) collecting in large volume of buffer ➡️ hard to concentrate for mass spec preps, and 2) we had to collect for HOURS to get enough for a decent MS run 😖 (although, that might be different now with newer instruments 🤔)
- I am deciding what to purchase / invest in for the lab - I mostly use Milteniy homogenization which I appreciate for reproducibility and recovery (it’s pretty good) but - the consumables are 💰💰😒 There is an aged nitrogen cavitation vessel that I can inherit, but I’ve never used one 🤨

- How feasible is it with tissues where you don’t have a lot of input? Ie how low can you go ? 🙃I had mixed experience with dounce homogenization - there have been problem with reproducibility from person to person, even with same settings, and the recovery is low. However with percol the purity is ⬆️

- Thank you - the final use is proteomics, so the goal is purity > functional integrity. antibodies have been useful in getting rid of contaminants (in case of brain - synapses) but there is still A LOT more proteins than I’d hope for. I think the biggest variability comes from homogenization
